Critical Role of Toll-Like Receptor 9 in Morphine and Mycobacterium tuberculosis–Induced Apoptosis in Mice

چکیده
BACKGROUND Although it is established that opioid and Mycobacterium tuberculosis are both public health problems, the mechanisms by which they affect lung functions remain elusive. METHODOLOGY/PRINCIPAL FINDINGS We report here that mice subjected to chronic morphine administration and M. tuberculosis infection exhibited significant apoptosis in the lung in wild type mice as demonstrated by the terminal deoxynucleotidyl transferase-mediated deoxyuridine triphosphate nick end labeling assay. Morphine and M. tuberculosis significantly induced the expression of Toll-like receptor 9 (TLR9), a key mediator of innate immunity and inflammation. Interestingly, deficiency in TLR9 significantly inhibited the morphine and M. tuberculosis induced apoptosis in the lung. In addition, chronic morphine treatment and M. tuberculosis infection enhanced the levels of cytokines (TNF-alpha, IL-1beta, and IL-6) in wild type mice, but not in TLR9 knockout (KO) mice. The bacterial load was much lower in TLR9 KO mice compared with that in wild type mice following morphine and M. tuberculosis treatment. Morphine alone did not alter the bacterial load in either wild type or TLR9 KO mice. Moreover, administration of morphine and M. tuberculosis decreased the levels of phosphorylation of Akt and GSK3beta in the wild type mice, but not in TLR9 KO mice, suggesting an involvement of Akt/GSK3beta in morphine and M. tuberculosis-mediated TLR9 signaling. Furthermore, administration of morphine and M. tuberculosis caused a dramatic decrease in Bcl-2 level but increase in Bax level in wild type mice, but not in TLR9 KO mice, indicating a role of Bcl-2 family in TLR9-mediated apoptosis in the lung following morphine and M. tuberculosis administration. CONCLUSIONS/SIGNIFICANCE These data reveal a role for TLR9 in the immune response to opioids during M. tuberculosis infection.

Silymarin inhibits Toll-like receptor 8 gene expression and apoptosis in Ramos cancer cell line
Objective: Silymarin is a herbal extract containing flavonolignans, and it has inhibitory effects against the growth of different cancer cell lines by inducing apoptosis. Toll-like receptors are suggested as a novel and attractive target to treat cancer. The current study aimed at examining the mechanism of silymarin-induced apoptosis in Ramos cells and investigating its effect...
متن کاملToll-Like Receptor 9 Is Required for Opioid-Induced Microglia Apoptosis
Opioids have been widely applied in clinics as one of the most potent pain relievers for centuries, but their abuse has deleterious physiological effects beyond addiction. However, the underlying mechanism by which microglia in response to opioids remains largely unknown. Here we show that morphine induces the expression of Toll-like receptor 9 (TLR9), a key mediator of innate immunity and infl...
متن کاملNMDA RECEPTOR ANTAGONISTS ATTENUATE TOLERANCE INDUCED BY MORPHINE AND NERVE LIGATION IN MICE
The effect of NMDA (N-methyl-D-aspartate) receptor antagonists on tolerance to morphine antinociception was investigated in mice. Daily subcutaneous administration of 50 mg/kg of morphine hydrochloride for three days induced tolerance to different (3,6 and 9 mg/kg) test doses of morphine. The tolerance obtained was decreased by pretreatment of animals with single or repeated doses of compe...
متن کاملارتباط پلیمورفیسمهای ژنتیکی پذیرنده شبه Toll 4 (TLR4) و استعداد ابتلا به سل ریوی
Background & Aim: Toll-like receptor 2 (TLR2) and Toll-like receptor 4 (TLR4) are the members of the Toll-like receptor family and play important roles in recognition and subsequent immune response against mycobacteria. Two functional polymorphisms for TLR2 (Arg677Trp, C à T Arg753Gln, G à A) and TLR4 (Asp299Gly, A à G Thr399Ile, C à T) genes have been associated with a negative influence on ...
متن کاملThe effect of CCK receptor agonists and antagonists in sciatic nerve-ligated mice
The effects of caerulin, a CCK receptor agonist, and proglumide, a receptor antagonist, on hyperalgesia induced by sciatic nerve ligation, was studied in mice. Tolerance to the morphine response was obtained 3,7, 14,21 and 28 days after unilateral sciatic nerve ligation. Maximum hyperalgesia was found 14 days after nerve ligation. Caerulin increased morphine antinociception in nerve-ligated ani...
